The sources of error in assessing the adhesiveness and specificity of adhesiveness of cells by use of reaggregation techniques are discussed in terms of the biophysics of such systems. A new technique for obtaining an absolute measurement of the adhesiveness of reaggregating cells is described: this technique avoids sources of error inherent in earlier techniques. The particular use of this technique to investigate whether or not specific adhesion occurs is described in theory. This new method does not require a means of recognizing the species or tissue type of individual cells. Experimental results using this technique to study the specificity of adhesion of mixtures of cells from the sponges Halichondria panicea Pallas, 1766, Suberites ficus Gray, 1806 and Microciona
fallax Bowerbank, 1866 are reported. No evidence of specificity of adhesion was found.
